Hydrophobic and hydrophilic cell surface (glyco)proteinic components of Candida albicans - 04/03/08

Objective |
Cell surface hydrophobicity (CSH) has been shown to influence adhésion and virulence of the opportunistic fungal pathogen Candida albicans . The aim of this work was to identify the potential cell surface (glyco)proteins supporting the relative CSH according to the morphological phase and culture conditions of Candida albicans .
Material and methods |
To characterize cell surface hydrophobic and hydrophilic components, Candida albicans Zymolyase crude soluble extracts of germ tubes and blastoconidia were fractionated by hydrophobic interaction chromatography. Prior to extraction, Candida cells were biotinylated in order to label cell surface proteins and glycoproteins. Proteinic and glycosylated compounds of the fractionated extracts were then tested by SDS-PAGE and concanavalin A staining, and probed with streptavidin peroxidase conjugate.
Results and conclusion |
For both morphological phases, cell surface hydrophilic fractions corresponded to highly glycosylated components of high molecular masses (over 94-kDa) and to a 34-kDa glycoprotein, whereas hydrophobic material was mainly represented by proteins with molecular masses between 10- to 94-kDa. Cell surface glycosylated high molecular mass components were more slightly hydrophobic for blastoconidia grown at 22 °C than for yeasts grown at 37 °C. Chromatography allowed identification of two peculiar cell surface germ tube specific components: a glycosylated polydispersed material of 110- to 220-kDa, and a 30-kDa proteinic components. These two hyphal phase specific components were characterized as being very hydrophobic.

Objectif |
L'hydrophobicité de surface de Candida albicans semble influencer l'adhérence et la virulence du champignon. Le but du travail présenté est d'identifier les différents (glyco)protéines pariétales de surface pouvant être impliquées dans les variations relatives de cette hydrophobicité en fonction du stade morphologique et les conditions de culture du champignon.
Matériel et méthodes |
Afin de caractériser les constituants de surface hydrophobes et hydrophiles de Candida albicans , les protéines et glycoprotéines pariétales de surface de blastospores et de tubes germinatifs ont été marquées par la biotine. Après extraction par la Zymolyase, elles ont été fractionnées par chromatographie d'interactions hydrophobes. Les fractions ont été analysées par SDS-PAGE, puis par affino-détection à l'aide de la concanavaline A, et de la streptavidine pour mettre en évidence les (glyco)protéines de surface.
Résultats et conclusion |
Les constituants de surface hydrophiles correspondent dans leur ensemble à des composants fortement glycosylés de masse moléculaire élevée (> 94 kDa), ainsi qu'à une glycoprotéine de 34 kDa. Les blastoconidies cultivées à 22 °C ou à 37 °C ne présentent pas de différences notables de leur composition, si ce n'est que les composants glycosylés de surface sont plus hydrophobes pour les levures cultivées à 22 °C. Les constituants hydrophobes sont représentés essentiellement par des protéines de bas poids moléculaires (10 à 94 kDa). Le fractionnement par chromatographie a permis d'identifier deux composants de surface spécifiques du tube germinatif. Il s'agit d'une glycoprotéine de masse moléculaire comprise entre 110 et 220 kDa et d'une protéine de 30 kDa. Ces deux composants présentent une forte hydrophobicité.
