Objective |
The objective of this work is to show the feasibility of manufacturing from a spray drying process particles containing immunoglobulin G capable of being administered by inhalation via a pressurized metered dose inhaler.
Methods |
Spray drying were made from aqueous solutions containing IgG and two types of excipients, mannitol and trehalose, with two ratios: 25% w/w and 75%w/w. The physicochemical and aerodynamic properties of the powders obtained were characterized just after manufacturing and after 1 month of storage at 40°C/75% RH according to criteria defined as needed to satisfy an inhaled formulation with a pressurized metered dose inhaler. Maintain of the biological activity and the structure of IgG after atomization was also tested by slot blot and circular dichroism.
Results |
All spray-dried powders presented a median diameter lower than 5μm. The powders atomized with trehalose showed a solid state more stable than those atomized with mannitol. All atomized powders were in the form of wrinkled particles regardless the nature and the ratios of excipients. The results showed that the aerosolisation properties were compliant with the target, independently of the excipient used at a ratio of 25% w/w IgG-excipient. Moreover, the addition of excipient during the atomization process the denaturation of IgG was limited.
Conclusion |
This study showed that the use of trehalose as excipient could satisfy the requirements of an inhaled formulation with a pressurized metered dose inhaler.

Objectif |
L’objectif de ce travail est de montrer la faisabilité de fabriquer, à partir d’un procédé d’atomisation, des particules contenant une immunoglobuline G (IgG humaine BRP) capables d’être administrées par voie inhalée par l’intermédiaire d’un aérosol doseur pressurisé.
Méthodes |
Des atomisations ont été réalisées à partir de solutions aqueuses contenant l’IgG et deux types d’excipient : le mannitol et le tréhalose avec deux proportions 25 % m/m et 75 % m/m. Les propriétés physicochimiques et aérodynamiques des poudres définies comme indispensables pour satisfaire une formulation inhalée avec un aérosol doseur pressurisé ont été déterminées juste après fabrication et après 1 mois de stockage à 40°C/75 % HR. Le maintien de l’activité biologique et de la structure de l’IgG après atomisation ont été testés par les méthodes Slot Blot et de dichroïsme circulaire.
Résultats |
Toutes les poudres atomisées présentaient un diamètre médian inférieur à 5μm. Les poudres atomisées avec le tréhalose montraient un état solide plus stable que celles atomisées avec le mannitol. Tous les atomisats se présentaient sous la forme de particules ridées indépendamment de la présence et du taux d’excipients. Les résultats ont montré que les propriétés d’aérosolisation étaient conformes aux objectifs indépendamment de l’excipient utilisé dans une proportion de 25 % m/m d’IgG/excipient. De plus, grâce à l’ajout d’excipient pendant le processus d’atomisation la dénaturation de l’IgG a été limitée.
Conclusion |
Cette étude a montré que l’utilisation du tréhalose comme excipient peut répondre aux critères préalablement établis pour prétendre satisfaire aux exigences d’une formulation inhalée administrée avec un aérosol doseur pressurisé.
