• | A great interest was carried for the analysis of morphine, a powerful narcotic analgesic opiate which use is prohibited during competitions. As sample preparation is a key step for the determination of drugs in biological samples, the aim of this work consists of the optimization of the urinary human sample pretreatment conditions before quantification by GC/MS. |
• | Our study includes first the optimization of liquid-liquid extraction method. The solvent mixture MTBE/isopropanol (4:1) was selected since it has improved morphine extraction from the urinary matrix allowing a gain of 50% when compared to that used in our routine laboratory. |
• | We have then focused on the enzymatic hydrolysis which morphine is released to its glucuronide conjugated form. A univariate study was conducted involving “amount of enzyme”, “incubation temperature” and “duration of hydrolysis”, in order to evaluate the influence of each of these operating variables on the ratio “Amorphine/AIS” response and to set their experimental fields. |
• | Based on these results, an experimental design was established using the Box-Behnken model to determine, by multivariate analysis, the optimal operating conditions maximizing the “Amophine/AIS” response. In the field of doping, a great interest is carried for the analysis of morphine, a powerful narcotic analgesic opiate which use is prohibited during competitions. In order to confirm the abnormal analytical result in our anti-doping laboratory, a sensitive and selective gas chromatography-mass spectrometry (GC-MS) method was performed for the quantification of urinary morphine. As sample preparation is a key step for the determination of drugs in biological samples, the aim of this work consists of the optimization of the urinary human sample pretreatment conditions before quantification by GC/MS. Enzymatic hydrolysis associated with liquid-liquid extraction constitute the major pre-treatment steps. Our study has first focused on the optimization of the extraction solvents then to enzymatic hydrolysis which morphine is released from its glucuronide conjugated form. Onboard premiums, a study involving the effect of “amount of enzyme”, “incubation temperature” and “duration of hydrolysis” was conducted. This univariate study has enabled us to evaluate the influence of each of these operating variables on the area ratio of morphine to the internal standard (Amorphine/AIS) response and to set the experimental fields for each one of them. Based on these results, an experimental design was established using the Box-Behnken model to determine, by multivariate analysis, the optimal operating conditions maximizing the “Amophine/AIS” response. After validation, the analysis of response surface makes it possible to set the optimum operating conditions, which the ratio “Amorphine/AIS” is maximized. The retained conditions for enzymatic hydrolysis are 160μl of Escherichia coli glucuronidase enzyme during 6hours of incubation at a temperature of 36°C. The solvent mixture Methyl-t-Butyl Ether/isopropanol (4:1, v/v) was selected since it has improved morphine extraction from the urinary matrix allowing a gain of 50% when compared to that used in our routine laboratory. Our developed extraction method can be successfully applied for our forensic anti-doping analysis of morphin in human sample urine.

Dans le domaine du dopage, l’analyse de la morphine, un puissant analgésique narcotique opiacé dont l’utilisation est prohibée durant les compétitions, présente un grand intérêt. Afin de confirmer le résultat analytique anormal dans notre laboratoire d’antidopage, une méthode sensible et sélective de chromatographie en phase gazeuse couplée à la spectrométrie de masse (GC-MS) a été utilisée pour la quantification de la morphine urinaire. Comme la préparation des échantillons étant une étape clé pour la détermination des médicaments dans les échantillons biologiques, le but de ce travail consiste à optimiser les conditions de prétraitement des échantillons urinaires humains avant leur quantification par GC/MS. L’hydrolyse enzymatique associée à l’extraction liquide-liquide constitue les principales étapes du prétraitement. Notre étude s’est d’abord concentrée sur l’optimisation des solvants d’extraction, puis sur l’hydrolyse enzymatique, par laquelle la morphine est libérée de sa forme glucuroconjuguée. Une étude faisant intervenir, à la fois, les facteurs, « quantité d’enzyme », « température d’incubation » et « durée d’hydrolyse » a été menée. Cette étude univariée nous a permis d’évaluer l’effet de chacun des facteurs sur la réponse « Aire de la morphine par rapport à l’Aire de l’étalon interne (Amorphine/AEI)", et de délimiter le domaine expérimental de chacun d’entre eux. Sur la base de ces résultats, un plan expérimental a été établi à l’aide du modèle de Box-Behnken afin de déterminer par analyse multivariée les conditions optimales optimisant la réponse « Amophine/AEI ». Après validation, l’étude des surfaces de réponse a permit de définir les conditions optimales pour lesquelles le rapport « Amophine/AEI » est maximisé. Les conditions retenues pour l’hydrolyse enzymatique sont 160μL de l’enzyme glucuronidase Escherichia coli pendant 6heures d’incubation à une température de 36°C. Le mélange de solvants méthyle-t-butyl ether/isopropanol (4:1, v/v) a été sélectionné car il nous a permis d’obtenir une meilleure extraction de la morphine de sa matrice urinaire, avec un gain de 50 % par rapport à celui utilisé en routine dans notre laboratoire. Notre méthode d’extraction développée peut être appliquée avec succès pour notre analyse médicolégale de la morphine dans les échantillons urinaires humains.
