Abstract |
Objectives |
The aim of the present study was to validate a simple, sensitive, HPLC method of analysis of doxazosin in human plasma with fluorescence detection.
Methods |
The validated method employed one-step direct protein precipitation with acetonitrile. Chromatographic separation was attained using a reverse-phase 250mm×4.6mm 5μ Hypersil® BDS C 18 column and the mobile phase consisted of 10mm sodium dihydrogen phosphate dihydrate (pH=3.0) and acetonitrile at a ratio of (65:35 v/v). The method was evaluated in terms of linearity, precision, accuracy, selectivity and stability as per standard guidelines. The total run time was about 4.5min which make this method suitable for high throughput analyses. This method was applied to the bioequivalence study of two doxazosin tablets in healthy human volunteers.
Results |
Good linear response was achieved over the range of 5.0–200ng/mL. The observed within- and between-day assay precision ranged from 0.64% to 14.73%; accuracy varied between 94.11% and 105%. The 90% confidence intervals for the ratio Cmax, and AUC 0-∞ of the test product over those of reference were within the acceptable range (0.8–1.25) for bioequivalence.
Conclusion |
The developed method was simple and could be applied to therapeutic drug monitoring of doxazosin.

Objectif |
Le but de la présente étude était de développer une méthode d’analyse par HPLC, simple et sensible, pour doser la doxazozine dans le plasme humain.
Méthode |
La séparation chromatographique a été obtenue en utilisant une colonne Hypersil® BDS C18 de 5μm en phase inverse de 250mm×4,6mm et la phase mobile était constituée de 10mm de dihydrogénophosphate de sodium dihydraté (pH=3,0) et d’acétonitrile à un rapport de (65/35 v/v). La méthode a été évaluée quantitativement en termes de linéarité, précision, exactitude, sélectivité et étude de stabilité conformément aux directives standard. La durée totale d’analyse était d’environ 4,5min, ce qui rend cette méthode adaptée aux analyses à haut débit. Cette méthode a été appliquée à l’étude de bioéquivalence de deux comprimés de doxazozine chez des volontaires sains.
Résultats |
Une bonne réponse linéaire a été obtenue dans la plage de 5,0 à 200ng/mL. La précision du dosage observée pendant et entre les jours variait de 0,64 % à 14,73 % ; la exactitude variait entre 94,11 % et 105 %. Les intervalles de confiance à 90 % pour le rapport Cmax et l’AUC 0-∞ du produit à tester par rapport à ceux de référence se situaient dans la plage acceptable (0,8–1,25) pour la bioequivalence.
Conclusion |
La méthode développée était simple et pouvait être appliquée au suivi thérapeutique de la doxazozine.
