Résumé |
L’espérance de vie est en constante augmentation dans les pays développés et tend à se rapprocher de la longévité maximale de l’espèce humaine. Il existe de nombreuses théories explicatives de ce phénomène de vieillissement au niveau cellulaire et moléculaire à travers différentes hypothèses génétiques ou non génétiques, notamment oxydatives et de glycation. Aucun processus biologique unique qui expliquerait l’origine et les conséquences du vieillissement n’a été décrit à ce jour. Alors que les théories stochastiques invoquent des événements isolés ponctuels aléatoires, comme les mutations de gènes, les théories systémiques invoquent des successions enchaînées d’événements. Dans les théories génétiques, on parle surtout de vieillissement programmé, d’une durée de vie programmée ou d’horloge biologique comme s’il existait un compteur interne à la cellule. Il semble qu’un tel compteur existe aux extrémités du chromosome sous forme du « télomère ». Les théories stochastiques attribuent un rôle prépondérant aux espèces réactives de l’oxygène qui provoquent des attaques des cibles cellulaires, induisant des dommages oxydatifs moléculaires cumulatifs responsables du vieillissement. Mais le vieillissement est un phénomène complexe et multifactoriel et de nombreuses autres théories sont présentées. Ce travail est introductif, à une nouvelle théorie uniciste qui sera décrite par le docteur Christophe de Jaeger dans un prochain article original.

The life expectation is in constant increase in the developed countries and tends to get closer to the maximal longevity of the human race. Of numerous explanatory theories of this phenomenon of ageing at the cellular and molecular level through different hypothesis genetic or not genetic, in particular oxydatives and glycation. There is probably no unique biological process which would explain the origin and the consequences of the ageing. While the stochastic theories call upon unpredictable punctual isolated events, as the transfers of genes, the systematic theories call upon successions chained by events. In the genetic theories, we speak especially about programmed ageing, about a scheduled life expectancy or about a clock biological as if there was an internal meter in the cell. It seems that such a meter exists in the extremities of the chromosome in the form of the “telomere”. Rather, aging occurs because of the extensive absence of adaptive genomic information required for survival to, and function at, later adult ages, due to the declining forces of natural selection during adult life. Among the aging theories, stochastic theories confer a key role to the reactive oxygen species leading to molecular cumulative oxidative damages to cell targets responsible for aging. Thus, an oxidative stress occurs during aging; this oxidative stress is defined as an imbalance between the production of reactive oxygen species and the antioxidant defences (enzymatic and non enzymatic systems). But the ageing is a complex and multifactorial phenomenon and numerous other theories are presented. This present work is an introduction to a new unicist theory of aging that is going to be presented by doctor Christophe de Jaeger in a next original article.
