Introduction |
The aim of this study is the evaluation of Quantitative Microsphere System (QMS®) lamotrigine reagent performances on CDx90® (ThermoFisher Scientific).
Patients and methods |
The QMS® technique is based on a competitive inhibition of turbidity in presence of free lamotrigine. On the CDx90®, calibration is carried out in six points from 0 to 40mg/L with detection at 700nm (end point or kinetic). Serum samples from 17 epileptic patients were compared with an HPLC technique. Statistics were performed with Methval® software (P<0.05).
Results |
With the kinetic mode, limit of quantification is 0.70mg/L. Between run, precision (n=15) is 4.4% (2mg/L), 4.0% (15mg/L) and 6.4% (25mg/L). There is no correlation with time up to 15mg/L (r2=0.060) and bias is less than −7.1% for at least 16 weeks. Patient results (0.83 to 13.88mg/L) correlate with HPLC (r2=0.984 ; P<0.05) with a mean negative bias of −0.9mg/L (or −9.1%) (P<0.05).
Conclusion |
The QMS® lamotrigine reagent on CDx90® permits a fast (15min) and easy determination of this molecule in serum within the therapeutic range in epilepsy (2.5–15mg/L).

Introduction |
Cette étude a pour objectif l’évaluation des performances du réactif Quantitative Microsphere System (QMS®) lamotrigine sur le CDx90® (ThermoFisher Scientific).
Patients et méthodes |
La technique QMS® est basée sur une inhibition compétitive de la turbidité en présence de lamotrigine libre. Sur CDx90®, le calibrage est effectué en six points (0 à 40mg/L) avec lecture de l’absorbance à 700nm (point final ou cinétique). Les résultats de 17 patients épileptiques ont été comparés à ceux d’une technique de chromatographie en phase liquide haute performance (CLHP). Les statistiques ont été effectuées à l’aide du logiciel Methval® (p<0,05).
Résultats |
En mode cinétique, la limite de quantification est égale à 0,70mg/L. La reproductibilité (n=15) est égale à 4,4 % (2mg/L), 4,0 % (15mg/L) et 6,4 % (25mg/L). Il n’existe pas de corrélation entre le temps et les contrôles jusqu’à 15mg/L (r2=0,060) ; le biais est inférieur à −7,1 % pendant au moins 16 semaines. Les résultats de lamotrigine des patients sur le CDx90 (0,83–13,88mg/L) corrèlent avec la CLHP (r2=0,984 ; p<0,05), avec un biais moyen de −0,9mg/L (soit −9,1 %) (p<0,05).
Conclusion |
Le réactif QMS® lamotrigine sur CDx90® permet un dosage rapide (15min) et aisé de ce médicament dans la zone thérapeutique en épilepsie (2,5–15mg/L).
