Résumé |
L’allergie aux protéines du lait de vache (APLV) toucherait 3 à 6 % des enfants de moins de deux ans. Lorsque l’allaitement doit être interrompu, on fait appel à des substituts. Trois types de produits à antigénicité réduite doivent être distingués selon le degré d’hydrolyse, des formules partiellement hydrolysées (HA), extensivement hydrolysées, à base d’acides aminés. La potentialité d’un produit à prévenir ou traiter l’allergie ne peut être déterminée que par des études cliniques randomisées ayant une méthodologie correcte.

Étudier l’allergénicité d’un lait extensivement hydrolysé in vitro en chambre de Ussing, à partir d’un modèle animal (souris Balb/c) sensibilisé avec la protéine allergisante majeure du lait de vache la ß-lactoglobuline (ß-Lg).
Matériels et méthodes |
Soixante souris femelles Balb/c (22,50±0,27g) sont utilisées, 48 souris immunisées avec la (ß-Lg) native et 12 souris femelles Balb/c ne recevant aucun traitement (témoin). Des échantillons d’un lait extensivement hydrolysé, d’un lait standard infantile « premier âge » et d’un lactosérum bovin natif utilisés dans cette étude sont caractérisés sur le plan biochimique par un dosage des protéines totales par la méthode de Lowry. L’allergénicité des laits est étudiée, en chambre de Ussing, par des tests de provocation effectués sur des fragments jéjunaux de souris Balb/c sensibilisées par voie intra péritonéale à la ß-lactoglobuline (ß-Lg).
Résultats |
Les résultats obtenus montrent que la teneur en protéines des deux laits infantiles est comparable à celle du lait maternel. Sur le plan de l’allergénicité, le lait extensivement hydrolysé stimule significativement le courant de court-circuit (Isc), cette réaction est comparable à celle obtenue avec la protéine sensibilisante (ß-Lg), le lactosérum bovin natif, et le lait infantile standard. Ces résultats suggèrent l’existence d’une réaction anaphylactique locale due à l’interaction directe de l’antigène sensibilisant avec les muqueuses des fragments jéjunaux.
Conclusion |
Les peptides de faibles poids moléculaires du lait extensivement hydrolysé sont dotés d’un pouvoir allergénique chez les souris Balb/c.

The cow’s milk allergy touches 3 to 6% of the children less than two years. When the feeding must be interrupted, we appeal to substitutes. Three types of products in reduced antigenicity must be distinguished according to the degree of hydrolysis, partially hydrolyzed formulae, extensively hydrolyzed, with amino acids. The potentiality of a product to prevent or treat the allergy can be determined, by clinical randomized studies having a correct methodology.

Study of the allergenicity of the extensively hydrolyzed formula in vitro (Ussing chamber), from an animal model (Balb/c) mice, made sensitive with the major allergenic protein of cow’s milk (ß-Lg).
Materials and methods |
Sixty female mice Balb/c (22,50±0,27g), 48 mice immunized with native (ß-Lg) and 12 female mice Balb/c receiving no treatment.
Samples from, extensively hydrolyzed formula, standard formula “first age” and native bovine whey are used in this study and characterized on the biochemical plan by dosage of the total proteins (Lowry method). The allergenicity of formulae is studied in Ussing chamber, by tests of provocation done on jejunal fragments of Balb/c mice, sensitized by parenteral way with β-lactoglobulin (β-Lg).
Results |
The obtained results show that the amount of proteins of both, standard and extensively hydrolyzed formulae is close to the amount found in human milk. On the plan of the allergenicity, the extensively hydrolyzed formula stimulates the current of short circuit (Isc). This reaction is comparable to the one gotten with the sensitizing protein (ß-Lg), the native bovine whey, and the standard infantile milk. These results suggest the existence of a local anaphylactic reaction due to the direct interaction of the antigen making sensitive with the mucous membranes of jejunum fragments.
Conclusion |
The peptides of low molecular weights of the extensively hydrolyzed formula are endowed with an allergenic power to Balb/c mice.
