Relation entre les caractéristiques du mouvement des spermatozoïdes humains et leur fixation à la zone pellucide - 26/01/13

Introduction |
Notre objectif était d’étudier les relations entre les caractéristiques du mouvement des spermatozoïdes et leur fixation à la zone pellucide (ZP).
Patients et méthodes |
L’étude a porté sur 47 patientes candidates à une fécondation in vitro (FIV). La ponction des ovocytes a été effectuée sous anesthésie générale sous contrôle échographique après des cycles stimulés utilisant l’association soit le citrate de clomifène et le HMG ou la busereline et le HMG. L’étude de la mobilité des spermatozoïdes a été réalisée grâce à une videomicrographie assistée par ordinateur avant et après traitement du sperme au Percoll. La mise en contact des spermatozoïdes avec les ovocytes ayant donné des échecs de fécondation in vitro et préalablement conservés dans une solution saline a été réalisée pendant 90minutes à 37°C, air-CO2 à 5 %. Après incubation, les spermatozoïdes fixés à la ZP sont comptés grâce à un compteur à main.
Résultats |
L’étude du test de fixation des spermatozoïdes à la ZP a montré que l’interaction spermatozoïdes/ovocytes est corrélée positivement avec les paramètres de la mobilité analysée par videomicrographie assistée par ordinateur : la Vcl, la Vsl et l’ALH.
Conclusion |
La performance du test nécessite une maturation épididymaire préalable des spermatozoïdes en particulier l’ontogenèse des sites spermatiques de reconnaissance pour la ZP. Une altération de cette étape de maturation a abouti à des échecs de reconnaissance et de fixation à la ZP. Le test peut prédire le résultat de la FIV et orienter vers la bonne technique d’assistance médicale à la procréation (AMP).

Objective |
Our objective was to study the relationships between the characteristics of the movement of sperm and their attachment to the zona pellucida.
Materials and methods |
Study related to 47 patients candidates with an IVF protocol. The puncture of the ovocytes was carried out under general anesthesia under echography control after stimulated cycles using association is the citrate of clomifene and the HMG or the busereline and the HMG. Study of the mobility of the spermatozoa was carried out thanks to a videomicrography computer-assisted before and after treatment of sperm in Percoll. The setting in contact of the spermatozoa with the ovocytes having given failures of in vitro fertilization and beforehand preserved in a saline solution was carried out during 90minutes with 37°C, air-CO2 with 5%. After incubation, the spermatozoa fixed at the zona pellucida, are counted thanks to a meter with hand.
Results |
The study of the test of fixing of the spermatozoa at the zona pellucida showed that the interaction spermatozoa/ovocytes is correlated positively with the parameters of the mobility analyzed by videomicrography computer-assisted: Vcl, Vsl and the ALH.
Conclusion |
The performance of the test requires a preliminary epididymal maturation of the spermatozoa in particular the ontogenesis of the spermatic sites of recognition for the zona pellucida. A deterioration of this stage of maturation led to failures of recognition and fixing at the zona pellucida. The test can predict the outcome of IVF and towards the good technique of medical assistance to procreation (AMP).
