A simple, rapid, and sensitive RP-HPLC method using photodiode array detection was developed and validated for the simultaneous determination of butylhydroxyanisol and simvastatin with its impurities in tablet forms. Chromatographic separation was achieved on a Phenomenex Hypersil (250×4.6mm, 5μm) column using the mobile phase acetonitrile-sodium acetate (12mM) buffered to 4.2 with glacial acetic acid. The flow rate was 1.7mL/min, and the UV detection were made at 238nm for simvastatin and its impurities and at 290nm for butylhydroxyanisol. The system suitability solution used for peak impurity identification was generated in-situ without use of any impurity reference standard. The method was validated according to ICH Q2(R1) guidelines, and the acceptance criteria for accuracy, precision, linearity, specificity, robustness, LOD, and LOQ, were met in all cases. Moreover, the reproducibility results obtained by 22 Official Medicines Control Laboratories (OMCL) of European Directorate were satisfactory. The compounds selected for impurity validation were based on those found during long term and accelerate stability studies carried out on several formulation tablets from Moroccan and other markets. The described method was robust and successfully applied in quality control laboratories for routine analysis to determine the butylhydroxyanisol and simvastatin with its impurities content in tablet dosage forms.

Une méthode simple, rapide et sensible par chromatographie liquide haute performance couplée au détecteur à photodiode a été développée et validée pour la détermination simultanée du butylhydroxyanisole et de la simvastatine avec ses impuretés dans les comprimés. La séparation chromatographique a été effectuée au moyen d’une colonne Phenomenex Hypersil (250, 4,6mm, 5μm) en employant une phase mobile composée d’un mélange d’acétonitrile et d’acétate de sodium à 12mM, ajusté à pH 4,2, avec un débit de 1,7mL/min. La simvastatine et ses impuretés ont été détectées à 238nm et le butylhydroxyanisole a été détecté à 290nm. La solution du système de conformité utilisée pour l’identification des impuretés a été préparée en interne sans utilisation de standard de référence d’impureté. La méthode a été validée selon les guidelines ICH Q2(R1) ; et les critères d’acceptation en termes d’exactitude, fidélité, linéarité, spécificité, robustesse, LD, et LQ ont été respectés. En revanche, la reproductibilité des résultats obtenus par les 22 laboratoires officiels de contrôle des médicaments au niveau de la Direction européenne a été satisfaisante. Le choix des impuretés qui ont fait l’objet de la validation était basé sur les impuretés qui ont été formées durant la stabilité à long terme et accélérée des comprimés provenant des marchés Marocain et étrangers. La méthode présentée est robuste et appliquée avec succès en contrôle qualité de routine pour le dosage du butylhydroxyanisole et la simvastatine avec ses principales impuretés dans les comprimés.
