Abstract |
Purpose |
Breast angiosarcomas are rare vascular malignancies that arise secondary to irradiation or de novo as primary tumours. The aim of this study is to know whether c-myc amplification can reliably discriminate these two entities.
Materiel and methods |
Forty-seven patients treated for breast angiosarcomas were studied. Thirty-two patients were diagnosed with postradiation angiosarcomas after breast cancer treatment and 15 patients with primary angiosarcomas. Interphase fluorescence in situ hybridization (FISH) was performed by hybridization of probes covering C-MYC (chromosome 8q24.21) and CEP8 on tissue sections.
Results |
Amplification (5- to 20-fold) of the c-myc oncogene was found in all breast radiation-induced angiosarcomas (32 tumours) but in none of the 15 primary angiosarcomas except one (7%).
Conclusion |
This study reinforces that there are true pathogenetic differences between the two types of breast angiosarcomas which are morphologically indistinguishable. These data point the pathways preferentially involved in the pathogenesis of post radiation angiosarcomas of the breast and may provide the basis for an additional targeted therapy.

Objectif de l’étude |
Les angiosarcomes mammaires sont des tumeurs malignes vasculaires rares qui surviennent soit secondairement à une irradiation, soit de novo (angiosarcome primitif). L’objectif de cette étude était de savoir si l’amplification de c-myc permettait de distinguer ces deux entités.
Matériel et méthodes |
Les dossiers de 47 patientes prises en charge pour un angiosarcome mammaire ont été étudiés : 32 angiosarcomes en territoire irradié après traitement d’un carcinome mammaire et 15 angiosarcomes primitifs. La fluorescence in situ hybridization (FISH) a été réalisée sur coupes tissulaires avec des sondes c-myc (chromosome 8q24.21) et cep8.
Résultats |
Une amplification de c-myc (5 à 20 fois) était présente dans tous les angiosarcomes en tissus irradiés et dans un des 15 angiosarcomes primitifs (7 %).
Conclusion |
Cette étude confirme qu’il existe des différences pathogénétiques entre les deux types d’angiosarcome mammaire, qui ne peuvent pas être distingués morphologiquement. Ces données permettent également de préciser les voies de signalisation impliquées dans la pathogenèse des angiosarcomes mammaires qui pourraient être l’objet de thérapies ciblées.
