In vitro sensitivity of two different inocula of Microsporum canis versus terbinafine - 04/03/08

Purpose

Terbinafine is an allylamine derivative: it is the first antifungal agent with a primarily fungicidal action against dermatophytes. Its good efficacy in treating dermatophythosis has already been demonstrated both in humans and animals. In this study, the authors checked two type of inocula, non sporulating and sporulating mycelia of Microsporum canis , to evaluate the in vitro activity of terbinafine using a microdilution test.

Materials and methods

Twenty-nine unsporulated isolates and 18 sporulated samples of M. canis were tested to terbinafine. All the isolates were obtained from the coats of cats by brushing technique and seeded onto Mycobiotic agar. The strains were successively seeded into Tryptic Soy Broth (TSB) or onto Potato Carrot Agar (PCA) to obtain the inocula to assay the susceptibility of both mycelium or macroconidia. The assays were carried out by microdilution. All tests were performed in quadruplicate.

Results

All samples of unsporulated mycelia grew at concentrations of 0.005 mg/ml, 58 samples grew at concentrations of 0.025 mg/ml, 18 at 0.05 mg/ml. Growth at 0.5 mg/ml and higher concentrations was not observed. All samples of sporulating mycelia grew at 0.005 mg/ml, 24 grew at concentrations of 0.025 mg/ml, 20 at 0.05 mg/ml. No sample grew at concentrations of 0.5 mg/ml. When testing mycelia, a DE 50 of 0.03 mg/ml has been recorded, while 0.02 mg/ml was the DE 50 for the macroconidia.

Conclusions

Basing upon the statistical analysis of the data, no difference between the behavior of the two different types of inocula towards terbinafine was observed. The strong fungicidal character of terbinafine against M. canis has also been evidenced.

Sensibilitéin vitro à la terbinafine de deux inocula différents de Microsporum canis

Objectif

La terbinafine est un dérivé de l'allylamine douée d'une action fongicide contre les dermatophytes. Son efficacité dans le traitement des dermatophytoses a été demontrée aussi bien en médecine humaine qu'en médecine vétérinaire. Dans ce travail, l'activité in vitro de la terbinafine a été étudiée contre deux inocula différents de Microsporum canis constitués respectivement par du mycélium non sporulé et sporulé (macroconidies).

Matériels et méthodes

L'activité de la terbinafine comme médicament antifongique a été evaluée sur 29 inocula constitués de mycelium, et sur 18 inocula constitués de macroconidies de M. canis de provenance féline. Toutes les souches ont été ensemencées sur Mycobiotic agar. Toutes les souches ont été successivement ensemencées en Tryptic Soy Broth (TSB) pour le mycelium, et en PCA pour les macroconidies. L'activité de la terbinafine a été évaluée par un test de microdilution.

Résultats

Toutes les souches non sporulées se sont développées pour concentrations de terbinafine de 0,005 mg/ml, 58 pour des concentrations de 0,025 mg/ml, 18 pour des concentrations de 0,05 mg/ml. Aucune culture n'a été obtenue pour des concentrations de 0,5 mg/ml. Toutes les souches sporulées se sont developpées à concentrations de 0,005 mg/ml, 24 à concentrations de 0,025 mg/ml, 20 à concentrations de 0,05 mg/ml. Aucune culture n'a été obtenue pour des concentrations de 0,5 mg/ml. La DE 50 est de 0,03 mg/ml pour le mycelium, et de 0,02 mg/ml pour les conidies.

Conclusions

En analysant statistiquement les résultats obtenus, on peut affirmer que les deux inocula différents ont montré la même sensibilité à la terbinafine. On a aussi observé l'activité fongicide de la terbinafine sur les échantillons de M. canis examinés.
