Objectif |
L'action de cinq antifongiques systémiques a été étudiée comparativement sur l'activité déshydrogénase mitochondriale et sur les capacités d'adhérence et de filamentation de C. albicans .
Matériels et méthodes |
Deux modèles d'adhérence ont été développés : adhérence aux cellules épithéliales buccales et adhérence au polystyrène.
Résultats |
Les résultats ont montré que l'amphotéricine B, utilisée en concentration sub-inhibitrice, inhibait significativement l'ensemble des paramètres testés. En revanche, la 5-fluorocytosine, le kétoconazole, l'itraconazole et la terbinafine agissaient plus spécifiquement sur un ou plusieurs des paramètres testés, induisant soit leur augmentation (5-fluorocytosine, terbinafine), soit leur inhibition (kétoconazole, itraconazole, terbinafine). Il a été notamment observé que la terbinafine à concentration sub-inhibitrice n'agissait pas sur la filamentation, inhibait le métabolisme mitochondrial et l'adhérence au polystyrène, et à l'inverse favorisait l'adhérence aux cellules épithéliales buccales.
Conclusion |
Ce travail démontre l'importance du facteur concentration des antifongiques, qui en dessous d'un certain seuil, pourrait favoriser la colonisation fongique.

Objective |
The effects of five systemic antifungals were compared: their ability to affect mitochondrial dehydrogenase activity of Candida albicans was examined with their ability to affect adherence and filamentation of this yeast.
Materials and methods |
Two adherence models were developed: adherence to buccal epithelial cells and to polystyrene.
Results |
Results showed that amphotericin B, used at subinhibitory concentration, inhibited all tested factors. However, the action of 5-fluorocytosine, ketoconazole, itraconazole or terbinafine was more specific on one or a few tested factors, exhibiting either inhibition (ketoconazole, itraconazole, terbinafine) or improvement (5-fluorocytosine, terbinafine). It was in particular observed that a subinhibitory concentration of terbinafine did not affect filamentation, but inhibited mitochondrial metabolism and adherence to polystyrene, and on the contrary, improved adherence to buccal epithelial cells.
Conclusion |
Together these results highlighted the importance of the antifungal concentration which, below a certain threshold, could promote colonization of the host tissues by C. albicans .
