Effet synergique in vitro d'un mélange de latex d' Hevea brasiliensis et d'huile essentielle de Thymus vulgaris avec l'amphotéricine B sur la croissance de Candida albicans |
L'action d'un mélange de latex et d'huile essentielle de Thymus vulgaris a été testée sur la CMI 80 % de l'amphotéricine B dans une culture de Candida albicans ATCC90029 dont la croissance est suivie par spectrophotométrie. Pour cela la méthode de macrodilution avec traitement mathématique des données expérimentales, permettant une modélisation de la croissance, a été réalisée.
L'analyse de l'huile essentielle par chromatographie gazeuse et spectroscopie de masse (CG/SM) montre une forte teneur en thymol (51,26 %). Les tests antifongiques indiquent que le latex d' H. brasiliensis , l'huile essentielle de T. vulgaris et l'amphotéricine B agissent de façon synergique contre la croissance de C. albicans . La concentration en amphotéricine B nécessaire pour inhiber 80 % de la croissance de C. albicans peut être fortement diminuée (42 %-86 %) par l'utilisation combinée d'huile de thym et de latex d' H. brasiliensis . La plus faible CMI 80 % a été obtenue avec un mélange de latex d' H. brasiliensis (61 µg proteine ml –1 ) et d'huile essentielle de T. vulgaris (0,006 µl ml –1 ) correspondant à une synergie de l'ordre de 86 %. Ces résultats sont en faveur d'une utilisation éventuelle des latex et huiles essentielles dans les thérapies antimycosiques.
The antifungal activity of Hevea brasiliensis latex alone or in combination with antifungal drugs has been established for several years. However the combination of latex and essential oils in concomitance with antifungal drugs was not tested until now. Hevea brasiliensis latex, Thymus vulgaris (thyme) oil and amphotericin B were investigated, using a broth macrodilution method, alone and in combination, for antifungal activity against Candida albicans ATCC 90029. The essential oil was analyzed by chromatography/mass spectroscopy (GC/MS) that shows a high level of thymol (51.26%). Antifungal tests indicated that H. brasiliensis latex, thyme oil and amphotericin B act synergistically against C. albicans growth. The concentration of amphotericin B necessary to inhibit C. albicans growth with 80% efficacy could be strongly decreased (42%-86%) by the use of thyme oil and H. brasiliensis latex in combination. The lowest MIC 80% was obtained with a mixture of H. brasiliensis latex (61 µg protein ml –1 ) and T. vulgaris essential oil (0.006 µl ml –1 ) corresponding to a synergy of about 86%. These results support the idea that latex and essential oils may be used as pharmaceutical drugs.
