Summary |
Introduction |
The objective of this study was to verify the participation of the nitric oxide as mediator of the effects of the physical activity on the cortical and trabecular bone in the osteoporotic rats with the nitric oxide synthase inhibition.
Syntesis |
Thirty Wistar female rats divided into six groups were used: (1) sham-operated; (2) sedentary osteoporotic; (3) active osteoporotic; (4) sham-operated with L-NAME; (5) sedentary osteoporotic with L-NAME; and (6) active osteoporotic with L-NAME. The trabecular bone percentage in all of the bones and the cortical thickness of the vertebral segments were measured. The means were compared by SNK test.
Results |
The nitric oxide synthase inhibition reduced of the cortical thickness in cervical and thoracic vertebrae of the sham-operated rats (normal). The sedentary osteoporotic group presented decrease of the trabecular bone percentage in the whole skeleton. The administration of L-NAME in the sedentary osteoporotic group caused different effect: increased the trabecular bone percentage in the long bones, and in the thoracic and lumbar vertebrae, and reduced the cortical thickness of the vertebrae. The physical exercise increased the bone mass of the cortical and trabecular bone in osteoporotic rats. But L-NAME impeded the increase of trabecular and cortical bone promoted by the physical activity in the tibia and in the first thoracic vertebrae of the osteoporotic rats.
Conclusion |
The nitric oxide is a mediator of the beneficial effects of physical activity upon bone mass in rats with osteoporosis and the nitric oxide synthase inhibition promoted different effect between the cortical and trabecular bone.

Introduction |
Déterminer, à l’aide de l’histomorphométrie, la participation du monoxyde d’azote comme médiateur des effets de l’exercice physique, sur l’os cortical et trabéculaire chez la rate ostéoporotique, grâce à l’inhibition de la synthèse du monoxyde d’azote par le L-NAME.
Méthode |
L’étude a porté sur 30 rates femelles divisées en six groupes : (1) témoin ; (2) avec ostéoporose sans exercice physique ; (3) avec ostéoporose et exercice physique ; (4) témoin avec L-NAME ; (5) avec ostéoporose sans exercice physique avec L-NAME ; (6) avec ostéoporose et exercice physique avec L-NAME. Ont été mesurés le pourcentage d’os spongieux dans tous les os et l’épaisseur de la corticale des segments vertébraux. Les moyennes ont été comparées par le test de SNK.
Résultats |
L’inhibition de la synthèse du monoxyde d’azote a réduit l’épaisseur de la corticale dans les vertèbres cervicales et dorsales des rats témoins. Le groupe ostéoporose sans exercice physique a montré une baisse généralisée du pourcentage d’os trabéculaire. L’administration de L-NAME dans le groupe avec ostéoporose sans exercice physique a provoqué une augmentation du pourcentage d’os spongieux dans les os longs et les vertèbres dorsales et lombaires et a réduit l’épaisseur de la corticale des vertèbres. L’exercice physique a augmenté la masse de l’os cortical et trabéculaire chez les rates ostéoporotiques. En revanche, le L-NAME associé à l’activité physique a inhibé l’augmentation d’os spongieux et cortical dans le tibia et les premières vertèbres dorsales chez les rates ostéoporotiques.
Conclusion |
Le monoxyde d’azote est le médiateur des effets bénéfiques de l’activité physique sur la masse osseuse chez les rates ostéoporotiques et l’inhibition de la synthèse du monoxyde nitrique exerce un effet différent entre l’os cortical et l’os trabéculaire.
