Résumé |
Le crizotinib, un inhibiteur de la tyrosine kinase du récepteur anaplastic lymphoma kinase (ALK), permet d'obtenir des taux de réponse jusque-là jamais atteint de 57 % à huit semaines au cours des cancers bronchiques non à petites cellules de stade métastatique présentant réarrangement pour le gène ALK. Avec de tels résultats, le crizotinib a fait l'objet d'une procédure accélérée aux états-Unis permettant l'obtention d'une Autorisation de mise sur le marché (AMM) sur des résultats d'une étude de phase I. Ces résultats doivent être confirmés avec une étude de phase II et deux études de phase III chez des patients dont la tumeur porte un réarrangement ALK. En France, la commission d'AMM a octroyé au crizotinib une Autorisation temporaire d'utilisation (ATU) de cohorte, le 15 décembre 2011 pour faciliter sa mise à disposition des patients en attendant l'AMM.

Crizotinib, an inhibitor of the receptor tyrosine kinase anaplastic lymphoma kinase (ALK), achieves response rates of 57 % at eight weeks in patients with stage IV non-small cell lung cancer with ALK rearrangements. With such results, the crizotinib followed an accelerated procedure in the United States and obtained the Food and Drug Administration (FDA) approval based on the results of phase I studies. The results should be confirmed with one phase II study and two phase III studies in patients with ALK rearrangements. In France, the Commission of Authorization for Marketing has granted an Authorization of Temporary Use (ATU) for cohort on the 15 December 2011 to allow its administration in patients before marketing authorization.
