Summary |
The present study reports the development and validation of a simple and rapid colorimetric method for chloroform (CHCl 3 ) detection in biological specimens, coupled with a dual-column (HS-GC-FID) confirmation method for quantitative analysis. The colorimetric method is based on a modified Fujiwara reaction yielding a characteristic pink coloration, with a detection limit of 0.1 mg/dL. For the HS-GC-FID method, samples were diluted with an aqueous n-propanol internal standard (IS) and analyzed using a dual capillary column with front and back detectors. The method exhibited linearity from 0.25 to 30 mg/dL for CHCl 3 with a LOD of 0.12 mg/dL, and a LOQ of 0.25 mg/dL. The accuracy (bias) range for CHCl 3 is –10.0% to 12.67, and the precision (%CV) ranges from 2.29 to 18.0%. No carryover was observed. Established methods were applied in a real homicidal forensic case study investigation. Colorimetric screening, indicated by the appearance of pink color, together with quantitative analysis of two case studies, confirmed chloroform at a lethal blood concentration of 24.2 and 25.7 mg/dL, establishing acute CHCl 3 poisoning as the cause of death. This study presents the development and application of a rapid, robust, and sensitive analytical methodology for the unequivocal confirmation of CHCl 3 .

La présente étude décrit le développement et la validation d’une méthode colorimétrique simple et rapide pour la détection du chloroforme (CHCl 3 ) dans des échantillons biologiques, couplée à une méthode de confirmation par chromatographie en phase gazeuse à double colonne (HS-GC-FID) pour l’analyse quantitative. La méthode colorimétrique repose sur une réaction de Fujiwara modifiée, produisant une coloration rose caractéristique, avec une limite de détection de 0,1 mg/dL. Pour la méthode HS-GC-FID, les échantillons ont été dilués avec une solution aqueuse de n-propanol comme étalon interne (EI) et analysés à l’aide d’une double colonne capillaire équipée de détecteurs en amont et en aval. La méthode a démontré une linéarité de 0,25 à 30 mg/dL pour le CHCl 3 , avec une limite de détection (LOD) de 0,12 mg/dL et une limite de quantification (LOQ) de 0,25 mg/dL. L’exactitude (biais) pour le CHCl 3 se situe entre –10,0 % et 12,67 %, et la précision (CV %) entre 2,29 et 18,0 %. Aucun effet de report n’a été observé. Des méthodes éprouvées ont été appliquées dans le cadre d’une enquête médico-légale sur un homicide. Le dépistage colorimétrique, révélé par l’apparition d’une coloration rose, associé à l’analyse quantitative de deux échantillons, a confirmé la présence de chloroforme à des concentrations sanguines létales de 24,2 et 25,7 mg/dL, établissant ainsi l’intoxication aiguë au CHCl 3 comme cause du décès. Cette étude présente le développement et l’application d’une méthode analytique rapide, robuste et sensible permettant la confirmation sans équivoque de la présence de CHCl 3 .
