Development and validation of an alternative method for radiochemical purity control of 68Ga-PSMA
Conclusion |

Objective |
Radiochemical purity control of gallium-68-labeled PSMA ( 68 Ga-PSMA) is a critical step prior to patient administration. Thin-layer chromatography methods described in the Summary of Product Characteristics rely on methanol, a toxic and problematic solvent. The aim of this study was to develop and validate a rapid, reliable, methanol-free alternative method.
Materials and methods |
The proposed method used an ITLC-SG support and a 0.5 M ammonium acetate mobile phase, without prior drying, over a distance of 5 cm. Validation followed the methodology of the French Society of Radiopharmacy (SoFRa).
Results |
The method separated 68 Ga-PSMA (Rf = 0.75–1) from radiochemical impurities (free and hydrolyzed/colloidal 68 Ga), which remained at the origin, with resolutions Rs = 2.0 and 2.4. Repeatability (CV[RCP] = 1.27%; Rf standard deviation = 0.04), intermediate precision (CV[RCP] = 0.31%), trueness (mean bias = −0.03%), and robustness (mean CV[RCP] = 0.77% across the three factors tested) were compliant with validation criteria. The method was significantly faster than the reference method (1.7 vs. 4.4 min; Student's t -test, P < 0.001).
Conclusion |
The method was validated according to the SoFRa methodology, with precision comparable to the reference method and the use of a less toxic solvent. It constitutes a reliable, more rapid, and safe alternative for 68 Ga-PSMA RCP control, compliant with current regulatory requirements.
