Association of miR-29, miR-155, MMP-9, and MT1-MMP expression with lateral incisor root resorption adjacent to unilaterally impacted maxillary canines: A CBCT-based cross-sectional split-mouth study - 29/10/26
, Seema Gupta 7, Manish Sharma 8Highlights |
• | GCF miR-29, miR-155, MMP-9 and MT1-MMP were higher at lateral incisors with CBCT-confirmed external root resorption beside unilateral impacted maxillary canines. |
• | Group-by-side interactions were significant for all four markers; the impacted-versus-control difference was larger when root resorption was present. |
• | A cross-sectional split-mouth design reduced participant-level biological variability. |
• | The side effect in teeth without radiographic resorption is compatible with local follicular or eruptive activity and is not specific to root resorption. |
Summary |
Background |
External root resorption of the maxillary lateral incisor is a complication of impacted maxillary canines. This study evaluated the association between gingival crevicular fluid (GCF) expression of microRNA (miR)-29, miR-155, matrix metalloproteinase-9 (MMP-9), membrane type 1 matrix metalloproteinase (MT1-MMP), and CBCT-confirmed lateral incisor root resorption.
Methods |
This cross-sectional split-mouth comparative study included 50 consecutive patients aged 15–30 years with unilaterally impacted maxillary canines who met predefined clinical, periodontal, and radiographic eligibility criteria and were recruited between June 2024 and April 2026. Participants were classified according to the presence or absence of CBCT-confirmed lateral incisor root resorption ( n = 25 per group). GCF samples were collected from the lateral incisors adjacent to the impacted canine and contralateral control teeth. miR-29 and miR-155 expression was quantified using quantitative real-time polymerase chain reaction, whereas MMP-9 and MT1-MMP concentrations were measured using enzyme-linked immunosorbent assay. Linear mixed-effects models were used to evaluate the effects of the root resorption status, tooth side, and their interactions.
Results |
Fifty participants were included, comprising 25 participants with CBCT-confirmed lateral incisor root resorption and 25 without root resorption. The expression of all four biomarkers was significantly higher in participants with root resorption than in those without root resorption. Significant group-by-side interactions were observed for miR-29 (β = 0.807; 95% confidence interval [CI], 0.611-1.003), miR-155 (β = 0.472; 95% CI, 0.307-0.638), MMP-9 (β = 29.46 pg/mL; 95% CI, 12.96-45.95), and MT1-MMP (β = 39.66 pg/mL; 95% CI, 29.88-49.44) (all P < 0.001).
Conclusions |
Within the limitations of this study, increased GCF expression of miR-29, miR-155, MMP-9, and MT1-MMP was associated with CBCT-confirmed lateral incisor root resorption adjacent to the impacted maxillary canines. Although longitudinal studies are required to establish their utility, these biomarkers may reflect inflammatory and extracellular matrix remodelling.
