Une chimiothérapie infusionnelle ambulatoire comportant du 5-FU, de l'acide folinique (FOL) et du carboplatine a été administrée chez 45 patients porteurs d'un CPNPC avancé. Les patients ont été répartis en 3 groupes dont les heures de pic d'administration ont varié de 8 h en 8 h (heures de référence avec pics 5-FU et FOL à 4 h et carboplatine à 16 h ; ± 8 h). Le programme de référence s'est avéré significativement mieux toléré (aux points de vue hématologique et mucite) ; cette étude a validé prospectivement le concept de la chronotolérance de cette association chimiothérapique.

Abstract |
An infusional ambulatory chronotherapy with 5-FU, folinic acid (FOL) and carboplatin was administered to 45 advanced NSCLC patients. The patients were randomised into 3 groups varying according to the time of administration peak of drugs (reference hours: 4 a.m. for 5 FU (and FOL); 4 p.m. for carboplatin; ±8 h). The reference schedule appeared best tolerated (regarding hematology and mucitis). This study has prospectively validated the chronotolerance concepts of this complex combination chemotherapy.
