Highlights

Tenoxicam (TN) is an anti-rheumatic drug recommended to treat inflammation at various body sites.
Span-60 is a non-ionic surfactant potentially used to form niosomes, that can entrap high amount of drug in lipidic bilayer.
Anti-inflammatory test was conducted using lipopolysaccharide-activated cultured Human leukemia monocytic (THP-1) cells.
The cumulative tenoxicam release was fitted in different in vitro kinetic models, including zero and first order, Higuchi and Hixon-Crowell model.
The investigation presents an industrially applicable solution to develop novel niosomal TN loaded gel.

Summary

Objective

This investigation aimed to explore the potential of non-ionic surfactant based niosomal vesicles encapsulating tenoxicam (TN; anti-rheumatic drug) for the treatment of rheumatic diseases.

Material and methods

Mechanical dispersion technique with controlled pressure was employed to prepare different niosomal formulations. The effects of different ratios of surfactant (span-60), lipid, and sodium deoxycholate on noisome's physicochemical properties have been examined. Moreover, inhibition of TNF-α in lipopolysaccharide-activated cultured Human leukemia monocytic (THP-1) cells were demonstrated to assess the in vitro inflammation profile. Finally, the optimized niosomal formulation (TN3) was prepared in gel matrix consist of carbopol 934 (termed as TN34) and stability was also tested at 4±2 ̊C, 25±2 ̊C, 37±2 ̊C and 45±2 ̊C for 6 months.

Results

The optimized niosomal formulation exhibited a small vesicle size (165±14nm) and high drug encapsulation (79.64±1.5%). Niosomal gel formulation TN34 showed pH (6.7), viscosity (6810±3.34 cps), spreadability (19.11±1.87gm.cm/sec) and also displayed sustained release pattern of drug release (98.16±0.07% TN released from gel matrix in 24h) in vitro release study. TN34 exhibited substantial anti-inflammatory response, with ∼75% inhibition of TNF-α in 48h. Stability investigation revealed that refrigerator temperature is most suitable for the storage of niosomal gel.

Conclusion

Transdermal niosomal formulation displayed promising potential in the treatment of rheumatic diseases.

Objectif

Cette étude visait à explorer le potentiel des vésicules niosomales à base de surfactant non ionique encapsulant le ténoxicam (TN ; médicament antirhumatismal) pour le traitement des maladies rhumatismales.

Matériel et méthodes

Une technique de dispersion mécanique avec pression contrôlée a été utilisée pour préparer différentes formulations de niosomes. Les effets de différents ratios de surfactant (span-60), de lipides et de désoxycholate de sodium sur les propriétés physicochimiques des substances nocives ont été examinés. De plus, il a été démontré que l’inhibition du TNF-α dans les cellules monocytaires de leucémie humaine (THP 1) cultivées activées par des lipopolysaccharides permettait d’évaluer le profil d’inflammation in vitro. Enfin, la formulation niosomale optimisée (TN3) a été préparée dans une matrice de gel constituée de carbopol 934 (appelée TN34) et la stabilité a également été testée à 4°±2°C, 25°±2°C, 37°±2°C et 45°±°2C pendant 6 mois.

Résultats

La formulation niosomale optimisée présentait une petite taille de vésicule (165±14nm) et une encapsulation élevée du médicament (79,64±1,5 %). La formulation de gel niosomal TN34 présentait un pH de 6,7, une viscosité de 6 810±3,34 cps, une aptitude à l’étalement de 19,11±1,87gm.cm/sec, et présentait également un modèle de libération prolongée du médicament (98,16±0,07 % de TN libéré de la matrice de gel en 24heures). Le TN34 a présenté une réponse anti-inflammatoire substantielle, avec une inhibition d’environ 75 % du TNF α en 48h. L’enquête sur la stabilité a révélé que la température du réfrigérateur est la plus appropriée pour le stockage du gel niosomal.

Conclusion

La formulation topique de niosomes a montré un potentiel prometteur dans le traitement des maladies rhumatismales.
